If so, where are they located? Are we able to choose where they are located?

as far as I know Customer.io uses Google Servers located in the US and in the EU. When setting up your account you can choose the region yourself (for free). Besides that they us as a default SMPT-Server Mailgun, also in the US and EU. You can find more information about that here.
